Background

As the global number of COVID-19 cases continues to increase, the world is facing a serious shortage of testing for SARS-CoV-2. WHO stressed that the capacity to detect SARS-CoV-2 should be rapidly enhanced to improve early diagnosis of COVID-19 pneumonia.

 

Pooled Sample Testing of SARS-CoV-2 

Currently there are two ways to perform sample pooling for SARS-CoV-2 testing: 1. Multi-sample pooling, which requires pooling the swab samples from multiple individuals into one collection tube and removing a specific volume from the tube for nucleic acid extraction; 2. Multi-sample mixing and extracting, which refers to collecting of individual’s samples to separate collection tubes, and then removing of equal volume of preservation solution from each tube  for the nucleic acid extraction. At present, the main problems of pooled sample testing are the lack of high-throughput nucleic acid extraction products for mass screening, and the limited sensitivity of the reagents which can easily lead to false negative results after the sample pooling.

Pooled sample testing can significantly improve testing efficiency and reduce testing costs. The Lancet, JAMA, PLoS One, Pathology and other journals have published studies confirming the feasibility and reliability of pooled sample testing for SARS-CoV-2 and recommending it for mass screening. China, the United States, South Korea and Germany have adopted pooled sample screening testing for SARS-CoV-2 for large populations, and have issued corresponding policies and technical guidelines successively.
